What is the difference between a vocal remover and lead and backing separation?

A vocal remover gives you two files: vocals and instrumental. Every harmony, double and ad-lib is inside that single vocals file. Lead and backing separation goes one step further and splits that vocals file into the main line and everything sung around it.

Why would I separate backing vocals before making an AI cover?

Because a voice model converts whatever it is given. If the harmonies are still in the file, they are converted too, and the target voice comes back singing its own harmony stack against itself. Separating the lead first means only the lead is converted and the original harmonies can be mixed back underneath.
